Isobolographic antinociception of non-steroidal anti-inflammatory drugs in rodent formalin orofacial pain

Diverse studies suggest that non-steroidal anti-inflammatory drugs (NSAIDs) induce antinociception through inhibition of cyclooxygenases. In this study was evaluated the effect of NSAIDs to induce antinociception either alone or in combination in the formalin orofacial test of rodent. Male mice were injected intraperitoneally with dexibuprofen, dexktoprofen, diclofenac meloxicam, metamizol, and piroxicam, alone in the formalin orofacial assay and a dose-response were analyzed to obtain the ED50 of each drugs. Administration of NSAIDs produced a dose-dependent antinociception in both phases of the assay with different potency. Then combinations of the cited NSAIDs were tested and analyzed by isobolographic analysis. The results demonstrate that the nocifensive response induced when dexketoprofen (DEX) , the dextrorotatory enantiomer of S(+) configuration of ketoprofen, combined with piroxicam, diclofenac, dexibuprofen, metamizol, and meloxicam was synergistic, either in Phase I or Phase II of the formalin orofacial mice assay. In conclusion, these data demonstrated that the NSAIDs administered alone or in combinations produce antinociception. These effects need be explained by other mechanisms of action of NSAIDs that the simple inhibition of COXs. The findings could may be relevant for help relief acute or chronic pain, such as migraine, post‐herpetic neuralgia and tooth pain.
